Silencing of Lysyl Oxidase RNA Interference Suppresses Breast Cancer Metastasis

چکیده
Breast cancer is one of the malignant tumors that occur most commonly in women. The incidence shows an increasing tendency every year that maybe associates with some wrong modern lifestyle (Loh et al., 2011; Sulaiman et al., 2011). The main reason for the treatment failure is the metastasis and invasiveness of the cancer. Therefore, it is very important to find the possible mechanism of the recurrence and the metastasis of breast cancer. Lysyl oxidase (LOX) is an amine oxidase that crosslinks collagens and elastins in the extracellular matrix. LOX controls both the structure and the tensile strength of ECM, and thus acts to preserve tissue integrity. Recent studies indicated that LOX promotes malignant progression in cancer (Peyrol et al., 1997; Woznick et al., 2005; Fong et al., 2007). However, the specific mechanism of LOX-mediated promotion of breast cancer metastasis is unclear. In our research, we investigated the possible mechanism of LOX gene affecting on the invasion and metastasis of breast cancer cells by RNA interference (RNAi). LOX-RNAi-LV was constructed to lower the LOX gene expression level in the MDA-MB-231 breast cancer cells, so that we could observe how LOX gene regulates metastatic colonization and growth of breast

Silencing of rhomboid domain containing 1 to inhibit the metastasis of human breast cancer cells in vitro
Objective(s): A growing body of evidence indicates that rhomboid domain containing 1 (RHBDD1) plays an important role in a variety of physiological and pathological processes, including tumorigenesis. We aimed to determine the function of RHBDD1 in breast cancer cells. Materials and Methods: In this study, we used the Oncomine™ database to determine the expression patterns of RHBDD1 in normal a...
متن کاملLysyl oxidase mediates hypoxic control of metastasis.
Hypoxic cancer cells pose a great challenge to the oncologist because they are especially aggressive, metastatic, and resistant to therapy. Recently, we showed that elevation of the extracellular matrix protein lysyl oxidase (LOX) correlates with metastatic disease and is essential for hypoxia-induced metastasis. In an orthotopic rodent model of breast cancer, a small-molecule or antibody inhib...
متن کاملSilencing of CXCR4 blocks breast cancer metastasis.
RNA interference technology, silencing targeted genes in mammalian cells, has become a powerful tool for studying gene function. For the first time in cancer research, we show that direct injection of a pool of naked small interfering RNA (siRNA) duplexes can prevent tumorigenesis in an animal model, suggesting a novel preventive and therapeutic strategy for cancer management. As a model system...
متن کاملBcr-abl Silencing by Specific Small-Interference RNA Expression Vector as a Potential Treatment for Chronic Myeloid Leukemia
Background: RNA interference (RNAi) is the mechanism of gene silencing-mediated messenger RNA degradation by small interference RNA (siRNA), which becomes a powerful tool for in vivo research, especially in the areas of cancer. In this research, the potential use of an expression vector as a specific siRNA producing tool for silencing of Bcr-abl in K562 cell line has been investigated. Methods:...
متن کاملLentiviral Vector Mediated Claudin1 Silencing Inhibits Epithelial to Mesenchymal Transition in Breast Cancer Cells
Breast cancer has a high incidence and mortality rate worldwide. Several viral vectors including lentiviral, adenoviral and adeno-associated viral vectors have been used in gene therapy for various forms of human cancer, and have shown promising effects in controlling tumor development. Claudin1 (CLDN1) is a member of the tetraspan transmembrane protein family that plays a major role in tight j...
